Whole cell lysates (15 µg) from NTERA-2, Daudi, and indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with 1.0 µg/mL (1:500 dilution) of purified anti-BCAT1 antibody (clone W18024A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker. Cell lines were loaded in order of descending BCAT1 expression levels source (Human Protein Atlas).

Description

BCAT1 is an aminotransferase that catalyzes the catabolism of the branched-chain amino acids leucine, isoleucine, and valine, making it an important regulator of the TCA cycle. BCAT1 also plays a role in cell cycle progression, differentiation, and apoptosis. Clinical disorders involved with mutations in the BCAT1 gene include hypervalinemia and hyperleucine-isoleucinemia.

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone failed to immunoprecipitate BCAT1 from NTERA-2 lysates.

This clone was tested for immunocytochemistry on NTERA-2 cells fixed with 4% PFA and permeabilized with either methanol or Triton X-100. Both methods resulted in dim staining of BCAT1. We therefore do not recommend the clone for this application.
