Whole cell extracts (15 µg protein) from the indicated cell lines, A549 (negative control), RPMI 8226, U-937, THP-1, Jurkat, and MOLT-4 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-c-Myb antibody (clone W16209A) overnight at 4°C.     Whole cell extracts (300 μg total protein) prepared from Jurkat cells were immunoprecipitated overnight with 2.5 μg of purified rat IgG2a, κ isotype ctrl antibody (Cat. No. 400501) or purified anti-c-Myb antibody (clone W16209A). The resulting IP fractions and whole cell extract input (5%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of W16209A. Lane M: Molecular weight marker.

Description

c-Myb is a proto-oncogene protein that functions as a transcription factor and a key regulator of cell proliferation and differentiation. As a DNA binding protein, it plays a role in regulating cell cycle and gene expression. c-Myb expression is elevated in hematopoietic progenitor cells and decreases following cell differentiation. Increased or aberrant expression of c-Myb as well as duplication and c-terminal deletion have been reported in various malignancies suggesting it plays a critical role in tumorigenesis.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes
  • This clone is predicted to recognize isoforms 1, 2, 4, 7, and 8 based off complete sequence homology of the immunizing peptide and the corresponding region in each isoform.
  • ICC test failed. The clone incorrectly stains the negative control in the cytosol.
