IHC staining with Purified anti-Calponin 1 (clone CALP) on formalin-fixed paraffin-embedded human uterus tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., 1X (Cat. No. 928502), the tissue was incubated without (panel A) and with (panel B) Purified anti-Calponin 1 (clone CALP) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged. Scale bar: 50 µm -
A-431 cells (low expression cell line, panels A and C) and Saos-2 cells (high expression cell line, panels B and D) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-Calponin 1 (clone CALP), followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801) (Panels A and C). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm -
Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-Calponin 1 (clone CALP) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG Antibody (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
U-2 OS cells (positive cell line, filled histogram) and HeLa cells (low-expressing cell line, open histogram) were fixed and permeabilized using Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) and intracellularly stained with Purified anti-Calponin 1 (clone CALP), or Purified Mouse IgG1, κ Isotype Control (Cat. No. 401402) (open histogram, dashed line is representative for both U-2 OS and HeLa cells).

Calponin 1, encoded by the CNN1 gene, is an actin filament-associated regulatory protein found primarily in smooth muscle but also in non-muscle cells. Calponin 1 plays a significant role in regulating the contractility and structural integrity of the cytoskeleton by binding to actin and actin-interacting proteins, modulating their interplay. For example, by binding to actin and inhibiting myosin light chain kinase Calponin 1 reduces the phosphorylation of myosin light chains, preventing excessive contraction and ensuring appropriate smooth muscle response to physiological stimuli. Furthermore, Calponin 1 has been implicated in various physiological functions beyond muscle contraction including vascular tone and wound healing, making it a key player in maintaining homeostasis in various tissues.

Each lot of this antibody is quality control tested by western blotting. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.03 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
- Application Notes
-
For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following:
- Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100
- Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol
- 100% ice-cold methanol only
For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803). Fixation/permeabilization with either True-Phos™ Perm Buffer (Cat. No. 425401) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) is not recommended.
