Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-E. coli RNA Sigma 70 (clone 2G10.recM1) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker

Description

Most prokaryotes such as E. coli express a single core RNA polymerase to transcribe RNA from DNA. RNA Sigma 70, also known as rpoD, is the major housekeeping member of the sigma factor subunit family which are required for core RNA polymerase to bind to promoters and initiate transcription. RNA sigma 70 is also required for the unwinding of DNA strands to allow access to RNA polymerase.
