Glycodelin, also known as progesterone-associated endometrial protein (PAEP), belongs to the lipocalin family and exists in four glycoforms. The glycodelin isoforms are primarily expressed in different reproductive tissues, such as glycodelin-S in male seminal vesicles, glycodelin-A (GdA) in the endometrium, glycodelin-F (GdF) in fallopian tubes/follicular fluid, and glycodelin-C (GdC) in cumulus cells. Glycodelin isoforms are involved in reproductive processes such as implantation, fertilization, and pregnancy maintenance. GdA plays a crucial role in differentiating natural killer (NK) cells to decidual NK cells and modulating the NK cells to prevent maternal immune rejection during embryo development. At the feto-maternal interface, GdA inhibits the proliferation of T-cells and induces their apoptosis through Th receptors. GdC is converted from GdA and GdF through glycosylation modification and facilitates sperm-zona pellucida binding. Recently, abnormal glycodelin expression has been observed in various cancers, including ovarian, breast, colon, and lung cancers. Mice xenograft models have revealed that glycodelin plays a role in tumor proliferation and invasion.
