Glycogen Synthase Kinase 3 Alpha (GSK-3α) is a serine/threonine protein kinase which mediates the control of several regulatory proteins involved in glucose homeostasis, including glycogen synthase, and transcription factors such as JUN. It also plays a role in the WNT and PI3K signaling pathways. GSK-3α contributes to insulin regulation of glycogen synthesis by phosphorylating and inhibiting glycogen synthase (GYS1 or GYS2) activity and mediates the development of insulin resistance. In Wnt signaling, GSK-3α regulates the level and transcriptional activity of nuclear CTNNB1/beta-catenin. GSK-3α is also reported to facilitate the generation of amyloid precursor protein (APP)-derived amyloid plaques found in Alzheimer disease. To date, there are six GSK-3α mutant mouse lines generated, these stains express various abnormal phenotypes in growth, size, mortality, aging, adipose morphology, homeostasis and metabolism, muscle and the nervous system.
