HIF1, hypoxia induced factor 1, is a heterodimeric basic helix-loop-helix transcription factor composed of HIF1α and HIF1β. Under normoxic conditions, HIF1α undergoes prolyl hydroxylation which leads to degradation of HIF1α by the proteasome. Oxygen-starvation inhibits prolyl hydroxylation and prevents proteasomal degradation of HIF1α, leading to stabilization of HIF1. HIF1α contains a nuclear localization sequence, and following nuclear translocation HIF1α binds to the core DNA sequence 5'-[AG]CGTG-3' within the hypoxia responsive element (HRE). HIF1 is a master regulator of the response to hypoxia and induces transcription of multiple genes including VEGF, glucose transporters, and proteins that aid in adaptation to hypoxic conditions. HIF1 protein is also involved in the regulation of angiogenesis, erythropoiesis, and energy metabolism.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.125 - 0.5 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

This antibody (clone W22058A) is not recommended for immunocytochemistry (ICC).
For IHC-P, antigen retrieval using Tris-EDTA pH 9.0 buffer (10mM Tris, 1mM EDTA) is recommended. Antigen retrieval with Sodium Citrate H.I.E.R. (Cat. No. 928502) is not recommended due to low signal. - RRID
