Whole cell extracts (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-HOXB4 (clone W22219B) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat. No. 643807) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

HOXB4 is a member of the Homeobox (HOX) gene family, which plays a role in regulating embryonic development and maintaining adult tissue homeostasis. HOXB4 is a sequence-specific transcription factor with a homeobox DNA-binding domain. In hematopoiesis, HOXB4 enhances the self-renewal capacity of hematopoietic stem cells (HSCs), which are responsible for the continuous production of blood cells. Overexpression of HOXB4 in HSCs has been shown to lead to an expansion of these stem cells without inducing leukemogenesis. By binding to specific DNA sequences, HOXB4 activates a network of target genes that promote stem cell proliferation and inhibit differentiation, thus maintaining the stem cell pool.

HOXB4 also plays a pivotal role in organogenesis, particularly in the development of the spinal cord and the patterning of the vertebral column. During embryonic development, HOXB4 is expressed in a specific spatial and temporal manner, contributing to the formation of structures along the anterior-posterior axis. Its expression helps determine the identity and positional information of various segments, ensuring that body parts develop in the correct locations. In vertebrates, this function is crucial for the proper segmentation and alignment of the spine, as well as for the formation of the nervous system. Furthermore, aberrations in HOXB4 expression or function have been associated with various pathological conditions, including cancers. Dysregulated HOXB4 expression can lead to the uncontrolled proliferation of cells, contributing to tumorigenesis.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is not suitable for use in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P) or immunocytochemistry (ICC).

This antibody does not cross react with mouse HOXB4 in Western blotting (WB).
