    Human paraffin-embedded breast tissue slices were prepared with a standard protocol of deparaffination and rehydration. Antigen retrieval was done with citrate buffer 1X (10 mM, pH 6.0) at 95°C for 40 minutes. The tissue was washed with PBS/ 0.05% Tween-20 twice for five minutes and blocked with 5% FBS and 0.2% gelatin for 30 minutes. Then, the tissue was stained with 5 µg/mL purified anti-human CD248 (Endosialin) (clone B1/35) and 5 µg/mL purified Keratin 14 polyclonal (clone Poly19053) at 4°C overnight. On the next day, the tissue was washed twice with PBS and stained with Alexa Fluor® 647 goat anti-mouse IgG (clone Poly4053) (red) and Alexa Fluor® 594 donkey anti-rabbit IgG (clone Poly4064) (green) for two hours at room temperature. The nuclei were counterstained with DAPI (blue). The image was captured with a 20X objective.

    Human bone marrow-derived mesenchymal stem cells were fixed with 1% paraformaldehyde (PFA) for 10 minutes and blocked with 5% FBS for 30 minutes. Then the cells were stained with 5 µg/mL purified anti-human CD248 (Endosialin) (clone B1/35) in blocking buffer overnight at 4°C. On the next day, the cells were washed twice with PBS and stained with 2.5 µg/mL Alexa Fluor® 594 goat anti-mouse IgG (clone Poly4053) (red) for two hours at room temperature. The nuclei were counterstained with DAPI (blue). The image was captured with a 40X objective.

Description

Endosialin, also known as Tumor Endothelial Marker 1 (TEM1), CD248 and CD164 sialomucin-like 1 (CD164L1), is classified as a C-type lectin-like protein and shares both sequence and structural 39% homology with thrombomodulin (CD141) and 33% homology to complement receptor C1qRp (CD93). CD248 is expressed on pericytes and fibroblasts during tissue development, tumor neovascularization and inflammation.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
