Human paraffin-embedded liver tissue slices were prepared with a standard protocol of deparaffination and rehydration. Antigen retrieval was done with citrate buffer 1X pH 6.0 at 95°C for 40 minutes. Tissue was washed with PBS/ 0.05% Tween-20 twice for five minutes, permerilized with 0.5% Triton X-100 for 10 minutes and blocked with 5% FBS and 0.2% gelatin for 30 minutes. Then, the tissue was stained with 10 µg/mL of purified anti-human Hemopexin (HPX) (clone A20115H) at 4°C overnight. On the next day, the tissue was washed twice with PBS and stained with Alexa Fluor® 594 Goat anti-mouse (clone Poly4053) (red) for two hours at room temperature. The nuclei were counterstained with DAPI (blue). The image was captured with a 10X objective.

Description

Hemopexin, also known as beta-1B-glycoprotein, haemopexin, Hpx, and Hx, is a glycoprotein that has a molecular mass of approximately 63 kD and belongs to the hemopexin family of proteins. Hemopexin is the plasma protein with the highest binding affinity for heme. When oxidized, hemoglobin that circulates alone in the plasma (also known as free hemoglobin) turns into methemoglobin. Methemoglobin then further dissociates into free heme alongside the globin chain. When further oxidization occurs, the free heme will turn into free met-heme. This will cause the hemopexin to bind to the free met-heme, forming a met-heme and hemopexin complex until it reaches a receptor on hepatocytes or macrophages within the spleen, liver, and bone marrow. Evidence suggests that brain and blood iron homeostasis is perturbed in Alzheimer’s diseases. Deficiency of iron loading onto its major transporter protein, transferrin, was observed in Alzheimer’s, which suggests further examination of peripheral iron/heme metabolism.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
