Whole cell extracts (15 μg protein) from untreated (-) or UV-treated (+) NIH/3T3 cells were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1.0 μg/mL (1:5000 dilution) of purified anti-Jun Phospho (Ser73) for 2 hours at room temperature (left). Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Equal c-Jun loading was confirmed by probing membranes with anti-pan c-Jun (right). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a protein loading control at a 1:10000 dilution. NIH/3T3 cells were treated with UV and rested for 35 minutes, fixed with 4% paraformaldehyde for 10 minutes, permeabilized with Triton X-100 for 10 minutes, and treated with lambda protein phosphatase (LPP) (right panel) or left untreated (left panel). Cells were then blocked with 5% FBS for 60 minutes and intracellularly stained with 5.0 µg/mL (1:100 dilution) of purified anti-Jun Phospho (Ser73) overnight at 4°C, followed by incubation with Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) at 2.5 µg/mL. Nuclei were counterstained with DAPI and the image was captured with a 60X objective. Scale bar: 50 µm -

c-Jun is the first oncogenic transcription factor ever discovered. It heterodimerizes with members of the Fos protein family, such as c-Fos, to form the AP-1 transcription factor complex, exhibiting both activating and repressive functions depending on the interacting partner and promoter sequence context. c-Jun is required for cell cycle progression, and is protective against DNA damage, such as that resulting from ultraviolet light exposure. c-Jun is also overexpressed in many cancers including lung, breast and vulvar cancer, and is a target for anti-cancer therapies. Phosphorylation of c-Jun at serine 63 and 73 is activational, and mediated by the MAPK signaling cascade.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 3.0 - 5.0 μg/mL is recommended. For intracellular flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
