Cell lysate from HepG2 (lane 1), A549 (lane 2), PC3 (lane 3), HaCaT (lane 4), MCF-7 (lane 5), HeLa (lane 6) and NIH3T3 cell lines (lane 7) (15 µg protein for each) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to nitrocellulose, and probed with 1 µg/mL (1:500 dilution) of purified anti-Mac-2 (Galectin-3) antibody (clone M1606B04) (upper). Proteins were visualized using chemiluminescence detection after incubation with a 1:3000 dilution of HRP conjugated anti-rat-IgG secondary antibody for the anti-Mac-2 (Galectin-3) antibody (upper panel) or a 1:5000 dilution of Direct-Blot™ HRP anti-β-Actin (clone 2F1-1, lower panel) antibody. Description

Galectin-3 (galactose-specific lectin 3) is a unique member of the β-galactoside-binding lectin family molecule, which plays an important role in regulation of cell proliferation, differentiation, and tumor cell metastasis. Galectin-3 is mainly found in the cytoplasm, but it is also seen in the nucleus and can be secreted by the non-classical, secretory pathway. In general, secreted galectin-3 mediates cell migration, cell adhesion, and cell-cell interactions through its high affinity binding with galactose-containing glycoproteins on the cell surface. Cytoplasmic galectin-3 exhibits anti-apoptotic activity and regulates several signal transduction pathways, whereas nuclear galectin-3 has been associated with pre-mRNA splicing and gene expression. Recent studies also indicate the potential role of galectin-3 as a biomarker for acute and chronic heart failure.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. For immunoprecipitation, the suggested use of this reagent is 1-5 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
