Total cell lysates (15 µg total protein) from MCF7, MOLT4 (negative/low expressers) and Daudi cells (positive control) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1 µg/mL of Purified anti-NLRP1 Antibody, clone 9F9B12, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

Description

NLRP1, also known as NALP1, is a member of the NOD-like receptor (NLR) family that is responsible for the recognition of pathogen-associated molecular patterns (PAMP). NLRP1 is a sensor component of inflammasome that is involved in innate immune response. NLRP1-dependent activation of caspase-1 drives rapid proteolytic activation of caspase-1 and the maturation of the proinflammatory cytokines IL-1β and IL-18. The activation of NLRP1 leads to cell death of macrophages via pyroptosis, which results in the release of cytosolic contents and induces inflammatory responses. NLRP1 is found to be expressed at high levels in the brain and plays a crucial role in neuronal cell death and neurodegenerative diseases.
