N-terminally HA-tagged PPH21 was immunoprecipitated with 12CA5 anti-HA antibody (cross linked to protein A sepharose) from lysates of wtBY4741 (left) or PPM1δ (right) (lacking the PP2A methyltransferase) yeast cells, separated by 10% SDS-PAGE and blotted onto nitrocellulose membrane. Blots were incubated at 4°C o/n in a Miniblotter 45 (Immunetics) with 2A10 at the indicated dilutions. Anti-HA tag clone 16B12 served as positive control.

Description

PP2AC (Serine/threonine-protein phosphatase 2A catalytic subunit alpha isoform), encoded by the PPP2CA gene, is the main phosphatase for microtubule-associated proteins (MAPs). PP2A can modulate the activity of phosphorylase B kinase casein kinase 2, mitogen-stimulated S6 kinase, and MAP-2 kinase. PP2AC is reversibly methyl esterified on Leu-309 by leucine carboxyl methyltransferase 1 (LCMT1) and protein phosphatase methylesterase 1 (PPME1). This carboxyl methylation influences the affinity of the catalytic subunit for the different regulatory subunits, which in turn, modulates the PP2A holoenzyme's enzyme activity and cellular localization.
