Peroxiredoxin 1, also called PRDX1, is one of six members of the superfamily of thiol-dependent peroxidases. Peroxiredoxin 1 can eliminate reactive oxygen and nitrogen species. As a critical antioxidant enzyme, it utilizes an N-terminal cystine to detoxify peroxides and protect cells from oxidative stress damage. As a sensor of inflammation response, extracellular Peroxiredoxin 1 is released to trigger TLR4-dependent secretion of TNF-α and IL-6 in macrophage and dendritic cell maturation. Peroxiredoxin 1 is increased in bone marrow-derived macrophages by lipopolysaccharide (LPS) and IFN-γ exposure. In microglia, the expression level of Peroxiredoxin 1 is regulated through ROS/p38/MAPK pathway and protects microglia from cell death. Overexpression of Peroxiredoxin 1 in rat primary hippocampus neuronal and PC12 cells promotes survival under amyloid β toxicity. In addition to its role as an antioxidant, oxidized Peroxiredoxin 1 forms an oligomer that acts as a chaperone to suppress the c-Myc oncogene and promotes tumor cell apoptosis.
