Total lysates (15µg protein) from HeLa (Lane 1), 20nM scrambled siRNA (lane 2) and 20nM SIRT5 siRNA (Lane 3) treated HeLa cells were resolved by electrophoresis, transferred to nitrocellulose, and probed with 1:1000 diluted (0.5µg/mL) purified anti-SIRT5 antibody (upper). Proteins were visualized using 1:3000 diluted HRP Goat anti-mouse IgG Antibody and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin Antibody (1:2000 diluted) was used as a loading control (lower). -
Total cell lysate (15µg protein) from THP-1, HeLa and Raw264.7 cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1:250 (2µg/mL), 1:1000 (0.5µg/mL) and 1:2500 (0.2µg/mL) diluted purified anti-SIRT5 antibody (clone O91G9) or competitor’s antibody used at manufactures recommended concentration (upper). Proteins were visualized by chemiluminescence detection using 1:3000 diluted HRP Goat anti-mouse IgG Antibody for clone O91G9 or 1:3000 diluted donkey anti-rabbit IgG Antibody conjugated to HRP for competitor’s antibody (upper). 1:2000 dilution of Direct-Blot™ HRP anti-β-actin antibody (clone 2F1-1) was used as a loading control (lower). Lane M: Molecular weight ladder, M* indicates longer exposure.

The seven mammalian Sirtuin isoforms are assigned to different Sirtuin classes on the basis of sequence homology. The mitochondrial SIRT5 belongs to class III. In the mitochondrial matrix, Sirt5 deacetylates and activates carbamoyl phosphate synthetase 1 (CPS1), which is increased when amino acid metabolism is upregulated during fasting. More importantly, SIRT5 regulates lysine succinylation and malonylation as evolutionarily conserved post-translation modifications. SIRT5 was proved to regulate many metabolic pathways, with fatty acid beta-oxidation and ketone body production as two pathways strongly enriched in lysine succinylation. Recently, Zhang et al demonstrated that SIRT5 was targeted to protein complexes on the inner mitochondrial membrane via affinity for cardiolipin in order to promote respiratory chain function.
