SET and MYND domain-containing protein 2, also known as SMYD2, is one of five members of the SMYD family of lysine methyltransferases composed of an evolutionarily conserved SET domain and a zinc finger-containing MYND domain. SMYD2 localizes to the nucleus and cytoplasm and methylates histones such as H2B, H3K4, H3K36, and H4 as well as non-histone substrates, notably p53 on lysine 370. As a protein lysine methyltransferase, SMYD2 is a regulator of transcription and epigenetic signaling and functions in the regulation of cell cycle and apoptosis. It has also been shown to promote NF-κB signaling pathways via methylation of TRAF2 and STAT3. Aberrant SMYD2 expression is associated with a variety of cancers and other diseases including kidney disease, cardiovascular disease, and rheumatoid arthritis. It promotes tumorigenesis and metastasis through the repression of tumor suppressor proteins RB1, p53, and PTEN.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.25 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
