A family of proteins that is structurally similar to the angiopoietins has been identified as angiopoietin-like proteins (ANGPTLs). To date, eight ANGPTLs have been discovered, namely ANGPTL1 to ANGPTL8. All ANGPTLs contain an amino-terminal coiled-coil domain, a linker region, and a carboxy-terminal fibrinogen-like domain. ANGPTLs have been generally considered orphan ligands as they do not bind to the receptors classically targeted by angiopoietins, namely the tyrosine kinase with immunoglobulin-like and EGF-like domain 1 (Tie1) and the endothelialspecific receptor tyrosine kinase (TEK or Tie2). ANGPTL3 is a main regulator of lipoprotein metabolism as it is directly involved in inhibition of lipoprotein lipase (LPL) activity. ANGPTL3 deficient mice exhibit low plasma HDL cholesterol and HDL phospholipid (PL). In humans, the complete absence of ANGPTL3 results in an increased LPL activity and mass and lower circulating free fatty acid levels. The physical association of ANGPTL3 with ANGPTL8 greatly increases its affinity for LPL and creates a very potent endocrine inhibitor of plasma triglyceride clearance in heart, skeletal muscle and brown adipose tissue. The identification of integrin αvβ3 as a receptor for ANGPTL3 on the vasculature further help in dissecting its role in lipid metabolism and inflammatory diseases. Additional functions of ANGPTL3 has been highlighted in literature as inducer of actin filament rearrangement in glomerular podocytes and be a promising non-invasive biomarker in diagnosis of chronic hepatitis.

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at - 20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
