Frizzled-5 (FZD5), a G-protein coupled receptor with seven transmembrane domains (7TM), is one member of ten Frizzled protein family. Frizzled proteins are the principal receptors for the Wnt family of ligands, mediating canonical Wnt signaling. Frizzled proteins are present in most of the animals, but not present in plants or in simpler (single cell) eukaryotes. In the presence of Wnt ligand, Wnt binds to extracellular cysteine rich domain (CRD) of Frizzled. A receptor complex forms between Frizzled and low-density lipoprotein receptor-related protein 5/6 (LRP5/6). Frizzled-5 is a receptor for WNT1, WNT3A, WNT5A, WNT7A, and WNT11. According to the identity of amino acid sequences, frizzled proteins can be divided into four subgroups: subfamily 1 (FZD1, FZD2, and FZD7), subfamily 2 (FZD5 and FZD8), subfamily 3 (FZD9 and FZD10), subfamily 4 (FZD4) and subfamily 5 (FZD3 and FZD 6). Human FZD5 shares about 95% amino acid sequence identity with mouse FZD5. FZD5 plays crucial roles during eye development and neuronal development. Loss of FZD5 in mice leads to progressive retinal degeneration. FZD5 is involved in the formation of neuron polarity. FZD5 is expressed early in cultured neurons at minor neurites and axonal growth cones.

- Molecular Mass
- The 451 amino acid recombinant protein has a predicted molecular mass of approximately 50.5 kD. The DTT-reduced and non-reduced proteins migrate at approximately 62 - 80 kD and 100 - 140 kD by SDS-PAGE respectively. The N-terminal amino acid is Ala.

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

- ED50 = 0.08 – 0.64 µg/mL as measured by its ability to inhibit Topflash reporter activity in HEK293T human embryonic kidney cells induced by 50 ng/mL of recombinant mouse Wnt3a.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
