    Mouse CD6 was aliquoted in PBS, pH 7.4 at 0.2 mg/ml. One aliquot was freeze and thawed four times (4x freeze/thaws), and compared to a control kept at 4°C (control). The samples were tested for their ability to support the adhesion of Jurkat cells.

Description

T-cell differentiation antigen CD6 is a type 1 transmembrane cell adhesion molecule that is a member of the scavenger receptor cysteine rich (SRCR) family of proteins. CD6 is composed of three scavenger receptor cysteine-rich (SRCR) extracellular domains, a transmembrane region, and a long cytoplasmic tail without intrinsic catalytic activity. It is expressed on peripheral blood T lymphocytes, thymocytes, subset of NK and B cells, and some hematopoietic cell precursors and brain cells. Soluble form of CD6 is detected in blood serum. CD6 has two ligands, CD166 (ALCAM) and CD318. It plays an important role in the stabilization and maturation of immunological synapses by interacting with ALCAM. Domain 3 of CD6 binds to ALCAM via the N-terminal domain while CD318 interacts with domain 1 of CD6. CD6 plays a role in cell to cell contacts, regulating T-cell responses via CD6-ALCAM interaction, as well as functioning as a costimulatory molecule promoting T-cell activation and T-cell proliferation by its involvement in the TCR/CD3 signaling cascade complex. CD6 has been shown to have a high binding affinity to highly conserved bacterial components such as lipopolylsaccharides (LPS) and Lipoteichoic acid (LTA), which allows it to help regulate inflammatory responses and secretion of cytokines in response to bacterial toxins. CD6 is also a popular target for the development of new therapeutic methods for autoimmune diseases such as multiple sclerosis due to its involvement in T-cell proliferation and activation.

Molecular Mass
The 624 amino acid recombinant protein has a predicted molecular mass of approximately 68.8 kD. The DTT-reduced protein migrates at approximately 90 - 100 kD and and non-reduced protein migrates at approximately 190 kD by SDS-PAGE. The predicted N-terminal amino acid is Leu.
Purity
>95%, as determined by Coomassie stained SDS-PAGE.

0.22 µm filtered protein solution is in PBS, pH 7.4
Endotoxin Level
Less than 0.1 EU per µg protein as determined by the LAL method.
Concentration
10 and 25 µg sizes are bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity
ED50= 0.15 – 0.6 µg/mL as measured by the ability of immobilized protein to support the adhesion of Jurkat cells. The Deep Blue Cell Viability™ Kit (Cat. No. 424702) was used measure the number of adherent cells.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency. Please conduct an experiment-specific validation to find the optimal ED50 for your system.
