HeLa cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with (A) Alexa Fluor® 594 Rat IgG2a, κ Isotype Ctrl Antibody (Negative, Cat. No. 400555) or (B-D) Alexa Fluor® 594 anti EIF4A1/DDX2A Antibody (Clone W17130C) overnight at 4°C. Nuclei were counterstained with DAPI (Blue, Cat. No. 422801). The image was captured with a 60X objective using KEYENCE BZ-X700 fluorescence microscope. Exposure time (Seconds) for (A-D) is 1/20. Concentrations for (A-B) is 0.4 µg/mL, (C) is 0.2 µg/mL and (D) is 0.1 µg/mL.

Description

EIF4A1 is an ATP-dependent RNA helicase. It can form the eIF4F complex, which is involved in translation initiation by binding with the 5' m7 GTP cap of the mRNA. It could unwind mRNA secondary structure at the 5’ UTR region and lead to the small ribosomal subunit binding and followed scanning of the initiator codon.
