Whole cell lysates (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-Pan AKT (clone A21001A).     MCF-7 cells (high expression control, top panels) and HEK293 cells (medium-low expression control, bottom, panels) were fixed and permeabilized with cold methanol, blocked with 5% FBS, and then stained with Purified anti-Pan AKT (clone A21001A) (middle and right panels) or Purified mouse IgG2b, κ Isotype Control (Cat. No 401201) (left panels), followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322). Nuclei were counterstained with DAPI (Cat. No. 422801) and merged in the left and middle panel. Images were captured on a Revvity Operetta CLS™ High Content Analysis System with a with a 63X objective. Scale bar: 50 µm.

    IHC staining with anti-Pan AKT (clone A21001A) was performed on formalin-fixed paraffin-embedded human testis tissue following antigen retrieval using Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704). The tissue sections were incubated without (panel A) or with anti-Pan AKT (clone A21001A) (panels B and C), followed by incubation with Alexa Fluor® 647 Goat anti-mouse (Cat. Description

The AKT family of proteins, also known as Protein Kinase B (PKB), consists of three isoforms: AKT1, AKT2, and AKT3. These serine/threonine kinases are pivotal in regulating various cellular processes, including glucose metabolism, apoptosis, cell proliferation, transcription, and cell migration. AKT1 is primarily involved in cellular survival pathways and is a key player in inhibiting apoptosis, promoting cell growth, and protein synthesis. AKT2 is crucial in the insulin signaling pathway, facilitating glucose transport and maintaining glucose homeostasis. AKT3 is predominantly expressed in the brain and is essential for brain development. Dysregulation of AKT signaling is associated with numerous diseases, including cancer, diabetes, and neurological disorders such as Alzheimer's disease. Overexpression or mutations in AKT genes can lead to uncontrolled cell growth and survival, contributing to tumorigenesis in various cancers. Additionally, aberrant AKT signaling is implicated in insulin resistance and type 2 diabetes, as well as neurodegenerative diseases.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For immunohistochemistry, a concentration range of 2.5 - 10.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody detects all 3 isoforms of AKT protein kinases (AKT1, AKT2, AKT3).

Additional Product Notes

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using either True-Phos™ Perm Buffer (Cat. No. 425401), or Cyto-Fast Fix/Perm Buffer set (Cat. No. 426803)

For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using either PFA-Triton X-100, or Methanol only.

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).
