CUL3 (Cullin-3) is a key component of the ubiquitin-proteasome system (UPS), an essential pathway for regulating protein degradation in human cells. It functions as part of an E3 ubiquitin ligase complex, which tags specific proteins with ubiquitin molecules, marking them for degradation by the proteasome. CUL3 is a member of the Cullin family of proteins, which provide a structural scaffold for the assembly of multi-subunit ubiquitin ligases, termed Cullin-RING ligases (CRLs). These ligases play a central role in maintaining protein homeostasis by controlling the turnover of numerous regulatory proteins involved in critical cellular processes such as cell cycle progression, signal transduction, transcription, and stress responses.
CUL3 forms a complex with a RING-domain protein, RBX1, which recruits the E2 ubiquitin-conjugating enzyme, and various substrate adaptors known as BTB domain-containing proteins. The BTB proteins determine substrate specificity, allowing CUL3 to target a wide range of proteins for degradation. This ability to selectively degrade proteins makes CUL3 a crucial regulator of several physiological and pathological processes.
