Whole cell lysates (15 µg protein) from HeLa, HEL, MOLT-4, and RAW264.7 cells were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to a nitrocellulose membrane, and probed with 0.5 µg/mL (1:1000 dilution) of purified anti-DDX5 antibody, clone 14B9A58. Proteins were visualized using chemiluminescence detection by incubating the membrane with a 1:3000 dilution of HRP goat anti-mouse-IgG secondary antibody (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH antibody (1:2000 dilution, Cat. No. 649203) was used as a loading control (lower). Lane M: Molecular Weight Marker. -
Total cell lysate from 293T cells (lane 1) or NIH-3T3 (lane 2) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal anti-DDX5 (clone 14B9A58) antibody. Proteins were visualized using an anti-mouse-IgG secondary conjugated to HRP and chemiluminescence detection. -
HeLa cells were stained with purified anti-DDX5 (14B9A58) antibody, followed by staining with DyLight™ 488 conjugated goat anti-mouse IgG (green) antibody and Alexa Fluor® 594 conjugated phalloidin (red). Nuclei were stained with DAPI (blue).

DDX5, also named RNA helicase p68, is a prototypic member of the DEAD (Asp-Glu-Ala-Asp) box family. DDX5 has been shown to be involved in the processes requiring the alteration of RNA structures, such as the maturation of pre-mRNA, rRNA, and miRNA. DDX5 also plays important helicase-independent roles in transcriptional regulation. DDX5 can act as a transcriptional coactivator of the androgen receptor (AR), estrogen receptor α (ERα), tumor suppressor p53, MyoD, RUNX2, and β-catenin, which are involved in the regulation of cell proliferation, differentiation, and tumorigenesis. DDX5 has been shown to be up-regulated in various cancers, including breast cancer, prostate cancer, and colorectal cancer, suggesting that DDX5 may participate in tumor development. DDX5 can also act as a transcriptional repressor through interacting with HDAC1.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 0.5 µg per ml. For immunocytochemistry, the suggested use of this reagent is 2.0 - 10 μg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
