Total cell lysates (15 µg total protein) from A431, NTERA2 (negative controls), Daudi, RPMI-8226 (positive controls) cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 0.5 µg/mL Purified anti-DEPTOR Antibody, clone W17128A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

DEPTOR/DEPDC6, found only in vertebrates, plays an important role in multiple cellular processes such as cell growth, autophagy, apoptosis, ER stress and adipogenesis. It inbibits mTORC1 and mTORC2 kinase activity through direct interaction with mTOR in these complexes. DEPTOR also associates with chromatin and may regulate transcription, and can act as either an oncogene or oncosuppressor depending on cell- or tissue-specific context.
