Total cell lysates (15 µg total protein) from HEK293T cells mock-transfected or transfected with a plasmid containing FLT4 were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-VEGFR-3 (FLT-4) Antibody, clone AFL4, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular Weight marker.

Description

VEGFR-3, also known as FLT4, is a member of the Tyr protein kinase family. The extracellular portion of VEGFR-3 contains 7 immunoglobulin (Ig)-like domains and the cytoplasmic portion contains a protein kinase domain. FLT4 regulates angiogenesis and lymphangiogenesis, its ligands are VEGF-C and D and its binding is mediated by the 2nd and 3rd Ig-like domains of FLT4. During fetal development VEGFR-3 is expressed on endothelial cells, however, in the adult mice, the vascular endothelial cells lose VEGFR-3 expression, but the lymphatic endothelium expresses it constitutively. In addition, VEGFR-3 expression can be induced in tumors with active angiogenesis.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Additional reported applications (for the relevant formats) include: ELISA1,2, immunoprecipitation1and Western blot1,2 from cell lysates, immunohistochemical staining1, 3 of frozen and paraffin embedded tissue sections and in whole-mount mouse embryos, in vivo blocking1, 5 of VEGF-C binding to inhibit angiogenesis, lymphangiogenesis, growth and metastasis of different tumor cells, in vitro blocking4 of VEGF-C binding to induce dispersion of endothelial cell clusters.

This clone detect multiple bands by western blot: a ~140 kD band corresponding to cleaved/soluble VEGFR-3, and 220 and 230 kD bands corresponding to unglycosylated and glycosylated full length VEGFR-3 (Bando, et al. 2004. Int J Canc. 111:184.)

This product has been reported by a collaborator to work for immunohistochemical staining of frozen tissue sections.
