Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.1 µg/mL of purified anti-PER1 antibody (clone W17208B) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker. -
Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.5 µg of purified rat IgG2a, κ isotype ctrl antibody (Cat. No. 400502) or purified anti-PER1 antibody (clone W17208B). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a mouse control antibody against a separate epitope of PER1, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Lane M: Molecular weight marker. -

PER1 (Period Circadian Regulator 1), a transcriptional repressor, is a component of the circadian clock and a member of the Period family. The circadian clock is the body’s internal time keeping mechanism, which regulates various physiological processes including hormone rhythms, locomotor activity, sleep cycle, and behavior. PER1 is expressed throughout peripheral tissues but expression is most significant in the suprachiasmatic nucleus (SCN) region of the brain which functions as the primary circadian pacemaker in mammals. PER1 is expressed in an oscillating rhythm with a period of about 24 hours which is maintained even in constant darkness. PER1 expression is light dependent, with exposure to light during subjective night increasing expression and shifting its phase in the SCN. Full length PER1 localizes to the cytoplasm with a shorter nuclear isoform localizing in the nucleus. Diseases associated with PER1 include periodic limb movement disorder (PLMD), advanced sleep phase syndrome, cardiovascular diseases and cancer.

For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.
