Whole cell extracts (15 µg protein) from serum-starved NK-92 cells untreated (-) or treated (+) with 100 ng/mL of recombinant human IL-2 (Cat. No. 589104) and 100 ng/mL of recombinant human IFN-α2 (Cat. No. 592702) for 20 minutes were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane and probed with 0.25 µg/mL (1:2000 dilution) purified anti-STAT4 Phospho (Tyr693) antibody (clone A19016A) for 2 hours at room temperature. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Equal STAT4 loading was confirmed using a pan STAT4 antibody. Lane M: Molecular weight marker. -
NK-92 cells untreated (negative control, open histogram), or treated with hIL-2 (100 ng/mL) and hIFNα2 (100 ng/mL) (positive control, solid histogram) were fixed and permeabilized using True Phos™ Perm Buffer (Cat No. 425401) and intracellularly stained with purified anti-Stat4 Phospho (Tyr693) (clone A19016A), or Mouse IgG1, κ isotype control (Cat No. 401401) (Open histogram, dashed line is representative for both, untreated and treated NK-92 cells) followed by PE-goat anti-mouse IgG (Cat No.405307). -
PBMC pre-treated overnight with cell activator cocktail (1:2500; Cat No. 423301), and then treated with (positive control, solid histogram) or without (negative control, open histogram) hIL-2 (100 ng/mL) and IFNα2 (100 ng/mL) for 30 min, fixed and permeabilized using True Phos™ Perm Buffer (Cat No. 425401) and intracellularly stained with Purified anti-Stat4 Phospho (Tyr693) (clone A19016A), or Mouse IgG1, κ isotype control (Cat No. 401401) (Open histogram, dashed line is representative for both, untreated and IL2/IFNα2 treated PBMC) followed by PE-goat-anti mouse IgG (Cat No. 405307).

The signal transducer and activator of transcription (STAT) family of transcription factors are DNA-binding proteins required for transcriptional regulation of several cellular and physiological processes in response to cytokine and growth factor stimulation. Inactive STAT proteins are present as cytosolic monomers. C-terminal tyrosine phosphorylation by upstream Janus kinases results in STAT dimerization, translocation to the nucleus, and increased affinity for response elements found in the regulatory promoter of STAT target genes.
In contrast to most other members of the STAT family, STAT4 expression is restricted to hematopoietic cells and responds to fewer cytokines, such as IL-12 and IL-2. STAT4, which requires phosphorylation at tyrosine 693 for activation, plays a pivotal role in the differentiation and proliferation of Th1 and Th17 cells, both of which are crucial effectors of chronic autoimmune disorders. In natural killer cells, STAT4 is required for normal development, maturation, and activation. Single nucleotide polymorphisms of STAT4 are implicated in the pathogenesis of systemic lupus erythematosis and rheumatoid arthritis, and are linked with increased risk of hepatocellular carcinoma.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For flow cytometric staining using our True-Phos™ Perm Buffer in Cell Suspensions Protocol, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

This antibody is predicted to recognize mouse STAT4 phosphorylated at tyrosine 694 and rat STAT4 phosphorylated at tyrosine 693 due to complete sequence homology between the immunizing peptide and the respective murine STAT4 orthologs.
