Total cell lysates (15 µg total protein) from A549, U87-MG, NTERA-2, and U937 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1:1000 dilution of Purified anti-Tubulin-γ Antibody, clone Poly6209, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP donkey anti-rabbit IgG Antibody (Cat. No. 406401) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

  • 2-g-tubulin
    Whole cell extract from A431 cells (Lane 1) and NIH3T3 cells (Lane 2) were resolved by electrophoresis, transferred to nitrocellulose and probed with rabbit anti-γ-tubulin antibody. Proteins were visualized using a donkey anti-rabbit secondary conjugated to HRP and a chemiluminescence detection system.

Description

γ-tubulin is a 51 kD member of the tubulin superfamily and is conserved between yeast and mammals. This cytoskeletal protein is highly expressed in centrosomes and is thought to be involved in cytoskeletal organization and mitosis. γ-tubulin is expressed at highest levels during prophase to metaphase and can be modified by phosphorylation. This protein has been shown to interact with α-tubulin, paxillin, and RAC GTPase activated protein 1. The Poly6209 antibody has been shown to be useful for Western blotting of human and mouse γ-tubulin.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is a dilution of 1:1000 - 1:8000. It is recommended that the reagent be titrated for optimal performance for each application.

This product may contain other non-IgG subtypes.

Additional Product Notes

The 50 µL size can be used for approximately 10 Western blots, and the 200 µL size can be used for approximately 40 Western blots.
