N-Cadherin (NCAD), also known as Cadherin-2 (CDH2), is a type I membrane protein that preferentially interacts in a homophilic manner. It is a member of the Cadherin super family, a family of calcium dependent cell adhesion molecules. N-Cadherin is primarily expressed in mesenchymal tissues such as neural tissue, retina, endothelial cells, fibroblasts, osteoblasts, mesothelium, myocytes, limb cartilage, oocytes, spermatids, and Sertoli cells. Human N-Cadherin is a 906 amino acid protein with a 25 amino acid signal peptide, a 134 amino acid N-terminal propeptide, a 565 amino acid extracellular domain, a 21 amino acid transmembrane domain, and a 161 amino acid cytoplasmic domain. N-Cadherin shares 98% amino acid sequence identity with both mouse and rat N-Cadherin within the extracellular domain. The N-Cadherin ectodomain can be cleaved by MMP24 which leads to the generation of a soluble 90 kD N-terminal soluble fragment and a 45 kD membrane-bound carboxy-terminal fragment 1. The Carboxy-terminal fragment is further cleaved by gamma-secretase into a 35 kD fragment. MMP24 mediated N-Cadherin cleavage in neural stem cells affects N-Cadherin mediated anchorage of neural stem cells to ependymocytes in the adult, leading to the modulation of neural stem cell quiescence. N-Cadherin is not only involved in cell-cell adhesion, but also in cell differentiation, embryogenesis, migration, invasion and signal transduction. Fibroblast growth factor receptor interaction with N-Cadherin has been implicated in contact-dependent survival of ovarian granulosa cells in a FGFR dependent manner. N-cadherin/FGFR also have a pivotal role in promoting metastasis through differential regulation of ERK and AKT. Due to N-Cadherin’s influence on cell migration and invasion, an upregulation of N-Cadherin has been shown to be common amongst aggressive cancer types. N-Cadherin promotes the survival of cancer cells by accelerating migration and invasion of cancer cells.

- Molecular Mass
- The recombinant protein consists of 804 amino acid mature form and 938 amino acid pro-protein with predicted molecular mass of approximately 88.6 kD and 103.9 kD respectively. The DTT-reduced proteins migrate at approximately 110 kD and 130 kD, and non-reduced proteins migrate at approximately 240-270 kD by SDS-PAGE. The predicted N-terminal amino acid is Asp and Ser.

- Concentration
- 10 and 25 µg sizes are bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
- Activity
- ED50 = 0.2 - 0.8 μg/mL as measured by the ability of immobilized protein to support the adhesion of U-118-MG cells. Calcein-AM (Cat. No. 425201) was used to measure the number of adherent cells.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
