IHC staining on formalin-fixed paraffin-embedded human brain. Tissues treated with (panel A) or without (panel B) the heating step for Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) were incubated with 2 µg/mL of Purified anti-GFAP (clone 2E1.E9) (Cat. No. 644702) primary antibody. Then, secondary staining was performed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) (magenta) for 1 hour at room temperature, and co-stained with 10 µg/mL of Alexa Fluor® 488 anti-Tubulin β 3 (TUBB3) (clone TUJ1) (Cat. No. 801203) (green). Nuclei were counterstained with DAPI (Cat. No. 422801) (blue). Images were captured with a 10X objective. Scale bar: 50 µm

Recommended Usage

Dilute to 1X with DI water before usage.

The optimal antigen unmasking condition is a function of temperature (90 to 100°C) and incubation time (5 - 30 minutes) and should be established by the end user.

Application Notes

For acidic antigen retrieval buffer (pH 6.0) refer to BioLegend’s Citrate Buffer,10X (Cat. No. 420902) and Sodium Citrate H.I.E.R., 1x (Cat. No. 928502)

For neutral antigen retrieval buffer (pH 7.6) refer to BioLegend’s Tris-Buffered Saline 20X (1.0 M) (Cat. No. 925601) and Tris-Buffered Saline 20X (1.0 M) with Tween 20 (Cat. No. 925501)
