Biotinylated Recombinant Human B7-H2 (ICOSL)-Fc chimera (Cat. No. 798704) (black circles) binds to immobilized human ICOS-Fc chimera (Cat. No. 599004). Ultra-LEAF™ purified anti-human CD275 (B7-H2, ICOSL) (clone W21036I) (purple squares) inhibits the binding in a dose-dependent manner, whereas the Ultra-LEAF™ purified rat IgG2a, κ isotype control (Cat. No. 400544) (gray triangles) does not have this effect. ND50 range: 0.01 - 0.07 µg/mL.

B7-H2, a member of the B7 family and the immunoglobulin superfamily, is a 40 kD protein also known as B7RP-1, B7h, B7-H2, GL50 and ICOS Ligand. Human B7-H2 is expressed by B lymphocytes, activated monocytes/macrophages, and dendritic cells. B7-H2 binds to a CD28-like receptor, inducible costimulator molecule (ICOS, AILIM, CRP-1), which is expressed by activated T cells. The interaction of ICOS with B7-H2 plays an important role in the T cell costimulation pathway.

Each lot of this antibody is quality control tested by blocking the binding of 0.04 μg/mL Biotinylated Recombinant Human B7-H2 (ICOSL)-Fc chimera to 1 μg/mL immobilized human ICOS-Fc chimera. ND50 range: 0.01-0.07 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
