Untouched CD8+ T cells were isolated from fresh human whole blood PBMCs using the MojoSort on Columns™ - Human CD8 T Cell Isolation Kit with BioLegend’s Large Columns (Cat. No. 480178). The negatively selected cells were then stained with PE anti-human CD8a (clone RPA-T8) (Cat. No. 301008) and APC anti-human CD3 (clone UCHT1) (Cat. No. 300412). The cells were then analyzed by flow cytometry gated on live cells (using Helix NP™ Blue) and CD45+ cells. -

The negatively selected cells were then stained with APC anti-human CD8a (clone RPA-T8) (Cat. No. 301014) and PE anti-human CD3 (clone UCHT1) (Cat. No. 300408). The cells were then analyzed by flow cytometry gated on live cells (using Helix NP™ Blue) and CD45+ cells.

MojoSort on Columns™ is BioLegend’s product line for column-based cell separation. This portfolio offers total solutions for column system setup as well as column-optimized formulations for reagents and kits.
This kit is designed for negative isolation of human CD8+ T cells from PBMCs harvested from whole blood and leukopaks. Non CD8+ cells are magnetically labeled by incubating your sample with the biotin antibody cocktail followed by magnetic Streptavidin Nanobeads. Once applied to our MojoSort on Columns™ System, the magnetically labeled cells are retained in the column matrix, while the untouched CD8+ T cells flow through the column and are collected into a clean tube. The depleted/labeled non CD8+ cells can be eluted from the column matrix for use as a control sample.

This kit is designed for negative isolation of CD8+ T cells from human PBMCs derived from whole blood or leukopaks. This kit is optimized for use with a column-based separation system such as MojoSort on Columns™ - Large Columns (Cat. No. 480178), Multistand (Cat. No. 480177) and Column Separator (Cat. No. 480179). This kit has also been tested for compatibility with columns from other suppliers. Please contact BioLegend Technical Service (tech-biolegend@revvity.com) for more information and guidance on compatibility with 3rd party columns and column systems. This product is designed research use only (RUO).
Important Note: This kit is not suitable for use with our handheld magnets. If you're using handheld magnets, we recommend the MojoSort™ Human CD8 T Cell Isolation Kit (Cat. No. 480011).
Test sizes/starting cell capacity:- One test is defined as cell separation from 1x107 human whole blood PBMCs or leukopak-derived PBMCs.
- The 100 tests size provides enough reagents for approximately 1x109 cells.
Each lot has been individually optimized. Do not mix and match components from different lots of different kits.
