Whole cell extracts (15 µg total protein per lane) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-ASCL1 (MASH1) (clone W23121G) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 420801) for 10 minutes, permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-ASCL1 (MASH1) (clone W23121G), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Description

ASCL1 (Achaete-Scute Family BHLH Transcription Factor 1), also known as MASH1, is a transcription factor that plays a crucial role in neurogenesis, particularly in the development of the nervous system. ASCL1 is a member of the basic helix-loop-helix (BHLH) transcription factor family and acts by binding to specific E-box sequences in the DNA to regulate the expression of target genes that are necessary for the cell fate determination, differentiation, and development. The protein is expressed in various tissues during development, including the developing spinal cord, cortex, and hindbrain. In the neural tube, ASCL1 influences the differentiation of neural progenitors into various types of neurons, including dopaminergic, serotonergic, and other interneurons. The loss of ASCL1 function during development can lead to neurological disorders such as Hirschsprung disease, congenital central hypoventilation syndrome, dysautonomia. ASCL1 has been found to be involved in the progression of certain cancers, such as lung and neuroendocrine tumors. In these tumors, ASCL1 is often overexpressed and contributes to tumorigenesis by promoting the survival and proliferation of cancer cells.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with either of the following:
-Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100
-Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol

For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Fixation/permeabilization with True-Phos™ Perm Buffer (Cat. No. 425401). Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) is not recommended.
