Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1 µg/mL Purified anti-Caldesmon (clone W22071A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Lane M: Molecular weight marker -
U-87 MG cells were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 5 µg/mL of Purified anti-Caldesmon (clone W22071A), followed by with Alexa Fluor® 594 Goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured by a Revvity Operetta CLS™ High Content Analysis System with a 60X objective. Scale bar: 50 µm -
IHC staining of Purified anti-Caldesmon (clone W22071A) on formalin-fixed paraffin-embedded human uterus (positive tissue, panel A and C) and skeletal muscle (negative tissue, panel B and D). Following antigen retrieval using Citrate buffer, 10X (Cat. No. 420902), the tissue was incubated with 5 µg/mL of Purified anti-Caldesmon (clone W22071A) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 10X (panel A and B) or 40X (panel C and D) objective. Scale bar: 50 µm

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

For immunohistochemistry (IHC-P), either Citrate Buffer, 10X (Cat. No, 420902) or Tris-EDTA pH 9.0 is recommended for antigen retrieval.
For immunocytochemistry (ICC), fixing with Fixation Buffer (Cat. No. 420801) is recommended. Alternatively, 4% paraformaldehyde fixation followed by either permeabilization with 0.5% Triton X-100 or ice-cold methanol is recommended.

Tissue distribution: high-molecular-weight Caldesmon (isoform 1) is predominantly expressed in smooth muscle; low-molecular-weight Caldesmon (isoforms 2, 3, 4 and 5) is widely distributed in non-muscle tissues and cells. Caldesmon is not expressed in skeletal muscle or heart.
