Adapter protein CRK, also known as CRK proto-oncogene adaptor protein or V-CRK avian sarcoma virus CT10 oncogene homolog, has two isoforms: CRKI (28 kD) and CRKII (40 kD). Closely associated is CRKL, or CRK-like protein (36 kD). CRKII and CRKL possess a C-terminal SH3 domain and a regulatory tyrosine phosphorylation site that are lacking in CRKI. CRK’s SH2 and SH3 domains serve as positive and negative regulators of transformation, respectively. CRK is vital to cell motility via activation of Rac1 by CRKII and CRKL and cell branching and adhesion via activation of Rap1 by CRKI. It is also vital to cell proliferation, apoptosis, formation and turnover of focal adhesions, PDGF-induced actin cytoskeleton rearrangement, cell migration, and transcription. Overexpressed in several cancers, CRK has been implicated in tumor proliferation, metastasis, and resistance to chemotherapy. This, in addition to its roles in natural killer cell activation and differentiation, lymphocyte regulation, B cell signaling, and T cell migration, makes CRK a valuable therapeutic target.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
