Total lysates (15 µg protein) from HeLa (human), U87-MG (human, low expression) and NIH3T3 (mouse) cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:2000 (0.25 µg/ml) purified anti-DDX17 antibody, clone 7G6B09. Proteins were visualized using chemiluminescence detection by incubation with HRP Goat anti-mouse secondary antibody (Cat. No. 405306, 1:3000 dilution). Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (Cat. No. 643807, 1:8000 dilution). -
HeLa cells were fixed with 2% paraformaldehyde (PFA) for 10 minutes, permeabilized with 0.5% Triton X-100 for five minutes, and blocked with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 2 µg/mL anti-DDX17 (p82, p72) Antibody (clone 7G6B09) followed by Alexa Fluor® 594 (red) conjugated goat anti-mouse IgG in blocking buffer for two hours at room temperature. Actin filaments were labeled with Alexa Fluor® 488 Phalloidin (green). Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective. -
Immunoprecipitation of DDX17 from Hela cell extracts. Lane 1 is 5% input. Immunoprecipitation was performed using protein G resins only (lane 2), mouse IgG isotype control (lane 3), and anti-DDX17 antibody (clone 7G6B09, lane 4). Western blot was performed using anti-DDX17 antibody (clone 7G6B09).

DDX17 is an ATP-dependent RNA helicase, belonging to the DEAD (Asp-Glu-Ala-Asp) box superfamily and highly related to the other key family member DDX5. The RNA helicase activity of DDX17 has been implicated in the regulation of RNA metabolism, such as RNA splicing, RNA editing, RNA degradation, and modulating secondary RNA structure. DDX17 can act as a transcriptional co-activator for the estrogen receptor ERα. It is also involved in muscle cell differentiation by co-activating MyoD. DDX17 regulates histone modification through interacting with the acetyltransferases p300, CBP, and PCAF, and the histone deacetylase HDAC1. DDX17 deficient mice exhibit severe developmental problems and die shortly after birth.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 0.5 µg per ml. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/ml is recommended. For immunoprecipitation, the suggested use of this reagent is 5.0 - 20 μg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
