Total cell lysates (15 µg protein) from HeLa, Jurkat and NIH3T3 were resolved by 4-20% Tris-Glycine gel electrophoresis, transferred to nitrocellulose, and probed with 1:500 diluted (1 µg/mL), 1:2000 diluted (0.25 µg/mL) and 1:5000 diluted (0.1 µg/mL) purified anti-Tata Binding Protein (clone 1TBP18) antibody (upper). Proteins were visualized by chemiluminescence detection using 1:3000 diluted HRP Goat anti-mouse IgG Antibody (Cat. No. 405306). Direct-Blot™ HRP anti-β-actin Antibody (Cat. No. 643807) was used as a loading control (lower). M: protein marker. -
Total lysates (15 µg protein) from 293T and 293T/Tata Binding Protein (TBP) CRISPR/Cas9 knockdown (KD) cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1:1000 diluted (0.5 µg/mL) purified anti-Tata Binding Protein antibody, clone 1TBP18 (upper). Proteins were visualized using chemiluminescence detection by incubating with 1:3000 dilution of HRP goat anti-mouse-IgG secondary antibody (Cat. No. 405306). 1:2000 dilution of Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 649203) was used as a loading control (lower). Lane M: MW ladder.

The TATA-binding protein, also known as TBP or Tata Binding Protein, is the general transcription factor that functions at the core of the DNA-binding multiprotein factor TFIID. Binding of TFIID to the TATA box is the initial transcriptional step of the pre-initiation complex (PIC), which plays a role in the activation of eukaryotic genes transcribed by RNA polymerase II. This protein is a component of the transcription factor SL1/TIF-IB complex, which is involved in the assembly of the PIC during RNA polymerase I-dependent transcription. The rate of PIC formation is assumed to be primarily dependent on the rate of the association of SL1 with the rDNA promoter. SL1 is involved in stabilization of nucleolar transcription factor 1/UBTF on rDNA.
