Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.125 µg/mL of Purified anti-TFAP2A (clone W20235D) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

    IHC staining of Purified anti-TFAP2A (clone W20235D) on formalin-fixed paraffin-embedded human skin tissue. Following antigen retrieval using 1X Citrate Buffer (Cat. No. 420902), the tissue was incubated with 0.25 µg/mL (panel A) of Purified anti-TFAP2A (clone W20235D), followed by incubation with 2.5 µg/mL of Alexa Fluor° 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801) (panel B). Images were merged of Purified anti-TFAP2A and DAPI (panel C).     Cells were fixed with Fixation Buffer (Cat. No. 420801) and permeabilized with 0.5% Triton X-100 for 10 minutes, and were subsequently blocked with 5% FBS for 1 hour at room temperature. Hep G2 cells (negative control, panel A and C) and HeLa cells (positive control, panel B and D) were then stained with 5 µg/mL of Purified anti-TFAP2A (clone W20235D), followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405422) for 1 hour at room temperature. Cells were also stained with Flash Phalloidin™ Green 488 (Cat. No. 424201) (green) and nuclei were counterstained with DAPI (Cat. No. 422801) (blue). The images were merged for both Hep G2 (panel C) and HeLa cells (panel D). The images were captured by a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 25 µm

Description

TFAP2A, or transcription factor AP-2-alpha, binds to a GC-rich DNA consensus sequence 5'-GCCNNNGGC-3'. Its functions are integral to numerous biological processes including embryonic ectodermal development, stem cell differentiation, and the regulation of cell growth and proliferation. TFAP2A is active in the neural crest and regulates a group of cells in the early embryo that give rise to many tissues and organs, including bones and other tissues of the head and neck. In humans, TFAP2A mutation causes Branchio-oculo-facial syndrome (BOFS). In kidney development, TFAP2A functions in distal nephron development dictating the progression of cells towards a fully differentiated state. TFAP2A is also involved in carcinogenesis and is highly expressed in various types of cancer, including bladder cancer, melanoma, head and neck squamous carcinoma, gastric cancer, ovarian cancer, lung adenocarcinoma, and nasopharyngeal carcinoma. In addition to its role in cancer, TFAP2A is a well-known regulator of craniofacial development and evolution.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.0625 - 0.5 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 0.25 - 1.0 µg/mL is suggested. For intracellular flow cytometric staining, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For use of this antibody (clone W20235D) in immunohistochemistry on formalin-fixed paraffin embedded tissue (IHC-P), antigen retrieval with Tris-EDTA pH 9.0 or Citrate Buffer, 10X (Cat. No. 420902) is recommended. 

For use of this antibody (clone W20235D) in immunocytochemistry (ICC), fixation with either Fixation Buffer (Cat. No. 420801) followed by permeabilization with 0.5% Triton-X or 100% ice-cold methanol is recommended.

For use of this antibody (clone W20235D) in intracellular flow cytometry (ICFC), fixation with Fixation Buffer (Cat. No. 420801) followed by True-Phos™ Perm Buffer (Cat. No. 425401) is recommended.

This antibody (clone W20235D) is not recommended for mouse or rat reactivity.
