Description

The c-Fos protein, or FBJ murine viral (v-Fos) oncogene homolog, is a member of the Fos family of transcription factors. C-Fos forms heterodimers with proteins of the Jun family of transcription factors to form the AP-1 (Activator Protein-1) complex, which regulates critical cellular processes, including cell proliferation, differentiation, and survival. C-Fos has been demonstrated to play a critical role in angiogenesis, bone development, and in the progression of cancer. C-Fos is also often used as an indirect marker of neuronal activity.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 µL per million cells in 100 µL staining volume or 5 µL per 100 µL of whole blood. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 488 has a maximum emission of 519 nm when it is excited at 488 nm.

Application Notes

For ICC we recommend PFA-MeoH fixation/permeabilization. We do not recommend methanol-only and PFA+ Triton X-100 fixation/permeabilization due to dim signal.

 

For ICFC, we recommend fixation/permeabilization with the True-Phos™ Perm Buffer. Cytokine method was tried and failed due to no signal.

Structure
c-Fos is a 330 amino acid protein with a predicted molecular weight of 41 kD.
Distribution

Expressed in the nucleus of many tissues/cells including placenta, brain, thyroid, central nervous system, lung, endometrium, and B lymphocytes

Function
FOS and JUN oncoproteins form dimeric complexes that stimulate transcription of genes containing AP-1 regulatory elements. FOS was the first transcription factor identified to have a critical function in the development of cells required for skeletal formation.
