C57 mouse frozen kidney section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature and blocked with 5% FBS for 1 hour at room temperature. Then the section was stained with 10 µg/mL of Alexa Fluor® 488 anti-mouse CD326/EpCAM (clone G8.8, green), 10 µg/mL of Alexa Fluor® 594 anti-mouse podoplanin (clone PMab-1, red) at 4°C overnight. Nuclei was counter stained with DAPI (blue). The image was scanned with a 10X object and stitched with MetaMorph® software.

    Formalin-fixed, 300 micron-thick mouse kidney section was blocked, permeabilized and stained overnight with CD31 (clone MEC13.3) Alexa Fluor® 647 (red) at 5 µg/mL, Tubulin Beta 3 (TUBB3)(clone AA10) Alexa Fluor® 488 (green) at 2.5 µg/mL, and Podoplanin (clone Pmab-1) Alexa Fluor® 594 (blue) at 5 µg/mL, optically cleared, then analyzed at 230 µm imaging depth on a confocal microscope.     Paraformaldehyde-fixed (4%), 500 μm-thick mouse kidney section was processed according to the Ce3DTM Tissue Clearing Kit protocol (cat. no. 427701). The section was costained with anti-mouse Podoplanin Antibody (clone PMab-1) Alexa Fluor® 594 at 5 µg/mL (green), and anti-mouse CD31 Antibody (clone MEC13.3) Alexa Fluor® 647 at 5 µg/mL (magenta) and counterstained with DAPI (blue). The section was then optically cleared and mounted in a sample chamber. The image was captured with a 20X objective using Zeiss 780 confocal microscope and processed by Imaris image analysis software.

Description

The mucin-type glycoprotein podoplanin is thought to be involved in the development of the lymphatic vascular system. Podoplanin is named after its expression in the kidney glomerular epithelial cells (podocytes). It has a potential role in tumor progression.

Recommended Usage

Each lot of this antibody is quality control tested by immunohistochemical staining on frozen tissue sections. For immunohistochemistry, a concentration range of 5.0 - 10 µg/mL is suggested. For 3D immunohistochemistry on formalin-fixed tissues, a concentration of 5.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 594 has an excitation maximum of 590 nm, and a maximum emission of 617 nm.

Function
Mediates effects on cell migration and adhesion through its different partners. During development plays a role in blood and lymphatic vessels separation by binding CLEC1B, triggering CLEC1B activation in platelets and leading to platelet activation and/or aggregation.
Interaction
Interacts with CLEC1B; the interaction is independent of CLEC1B glycosylation and activates CLEC1B; the interaction is dependent of sialic acid on O-glycans.
