    HEL cells were fixed with Fixation Buffer (Cat. No. 420801) and permeabilized with ice-cold methanol and then blocked with 5% FBS. Cells were then stained with Alexa Fluor® 647 mouse IgG2b, κ isotype control (Cat. No. 400330) (panel A) or Alexa Fluor® 647 anti-GATA1 (clone P84F5) (panel B) overnight. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm

Description

The GATA family of transcription factors is comprised of six members characterized by their binding to the consensus DNA sequence (T/A)GATA(A/G). GATA1, which was the initially described member of the GATA family, is predominantly expressed in cells of the myeloid lineage such as erythrocytes, eosinophils, mast cells, and megacaryocytes. Over the course of erythroid differentiation, GATA2 activates expression of the functionally distinct GATA1; promoter-occupied GATA2 is replaced/exchanged at certain promoters with the GATA1 transcription factor, a process known as the GATA switch. Genes induced by GATA1 include HBB, HBG1/2, ALAS2 and HMBS. Haploinsufficient heterozygous GATA1 knockout mice spontaneously develop erythroblastic leukemia, and GATA1-null mice die early in development due to profound anemia.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm.

Application Notes

For immunocytochemistry (ICC) use we recommend cell fixation/permeabilization with either PFA/Triton X-100 or PFA/methanol. Cell Fixation/permeabilization with methanol alone is not suitable for this antibody.

When using this clone for ICFC, we recommend using the True-Nuclear™ Transcription Factor Buffer Set.
