Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-AMD1 (clone W18041A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-β-actin (Cat. No. 664804) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining with Purified anti-AMD1 (clone W18041A) on formalin-fixed paraffin-embedded human testis tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated with (panel A) and without (panel B) Purified anti-AMD1 (clone W18041A) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged (panels C and D). Scale bar: 50 µm

AMD1 (Adenosylmethionine Decarboxylase 1) is an enzyme critical for polyamine biosynthesis, a process vital for cell growth, proliferation, and differentiation. It decarboxylates S-adenosylmethionine (SAM) to form decarboxylated SAM (dcSAM), a crucial step in the synthesis of spermidine and spermine, which plays a key role in stabilizing cellular structures like DNA and RNA.
AMD1 maintains normal cell function through polyamine regulation, while its dysregulation is associated with cancer, neurological diseases, and inflammation. In many cancers, such as colon, prostate, and breast cancer, AMD1 is overexpressed. This leads to elevated polyamine levels, promoting rapid cell division, uncontrolled growth, and tumor progression. Given its role in driving tumor growth, AMD1 is viewed as a potential target for cancer therapy. Inhibiting AMD1 may decrease polyamine production, thereby slowing cancer progression.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
It is not recommended using this product in immunocytochemistry (ICC) or intracellular flow cytometry (ICFC).
This antibody does not cross react with mouse AMD1 in Western Blot.
