Total cell lysates (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-Annexin A2 Antibody, clone W18057A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin Antibody (Cat. No. 643807) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular Weight marker. Cell lysates were loaded in order of ascending Annexin A2 expression; predicted expression data was obtained from Human Protein Atlas.

    HaCat cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with methanol for 10 minutes and blocked with 5% FBS for 30 minutes. Cells were then intracellularly stained with either Purified Rat IgG2a, κ Isotype Ctrl Antibody (Cat. No. 400502, panel A) or with purified anti-Annexin A2 Antibody, clone W18057A at 2.0 µg/mL (panel B, panel C without DAPI), overnight at 4°C, followed by incubation with Alexa Fluor® 594 Goat anti-rat IgG (Cat. No. 405422) at a 1:200 dilution. Nuclei were counterstained with DAPI and the image was captured with a 60x objective.

Description

Annexin A2 (ANXA2) is a member of the annexin family, which are calcium-dependent phospholipid-binding proteins that play a role in the regulation of cellular growth and signal transduction pathways. Annexin A2 forms a heterotetramer complex with p11(S100A10) protein on the plasma membrane which then co-localizes with CD44, tenascin-C, cathepsin B and tissue plasminogen activator (t-PA). This complex leads to the activation of growth factors, proteases, degradation of extracellular matrix (ECM) proteins, and results in an increase in cancer cell adhesion invasion, metastasis and tumor growth. Annexin A2 also increases cancer cell proliferation and cell survival via the MAPK/ERK1/2 pathway and involves in actin cytoskeletal rearrangement and regulates cancer cell migration.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 1.0 µg per ml. For immunocytochemistry, a concentration range of 1 - 5 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone failed to immunoprecipitate Annexin A2 from whole cell extracts prepared from HeLa cells.

Triton X-100 permeabilization failed to properly stain for ICC, methanol permeabilization is the preferred method.
