ICC, IHC-P - Verified - Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

For immunohistochemistry (IHC-P), either Citrate Buffer, 10X (Cat. No, 420902) or Tris-EDTA pH 9.0 is recommended for antigen retrieval.
For immunocytochemistry (ICC), fixing with Fixation Buffer (Cat. No. 420801) is recommended. Alternatively, 4% paraformaldehyde fixation followed by either permeabilization with 0.5% Triton X-100 or ice-cold methanol is recommended.

Tissue distribution: high-molecular-weight Caldesmon (isoform 1) is predominantly expressed in smooth muscle; low-molecular-weight Caldesmon (isoforms 2, 3, 4 and 5) is widely distributed in non-muscle tissues and cells. Caldesmon is not expressed in skeletal muscle or heart.
Cellular distribution: cytoskeleton, cytosol, and plasma membrane. - Function
- Regulates actomyosin interactions in smooth muscle and non-muscle cells which affects cell contractions; involved in cellular mitosis and in Schwann cell migration during peripheral nerve regeneration.
