IHC staining of Purified anti-CD14 (clone W23074F) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., 1X (Cat. No. 928502), the tissue was incubated without (panel A) and with (panel B) 10 µg/mL of Purified anti-CD14 (clone W23074F) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. The tissue was also co-stained with Purified anti-human CD21 (clone Bu32) (Cat. No. 354902) followed by incubation with Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature (panel C). Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged (panel D). Scale bar: 50 µm

Description

CD14 is a glycosylphosphatidylinositol (GPI)-linked membrane glycoprotein also known as LPS receptor. CD14 is expressed at high levels on monocytes and macrophages, and at lower levels on granulocytes. Some dendritic cell populations such as interfollicular dendritic cells, reticular dendritic cells, and Langerhans cells have also been reported to express CD14. It plays an important role in the innate immune system, and is the first pattern recognition receptor identified. CD14 primarily binds to bacterial lipopolysaccharide (LPS), a component of the outer membrane of Gram-negative bacteria. By binding to LPS, CD14 initiates a signaling cascade that leads to the clearance of pathogens.

IHC-P - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For use of this antibody (clone W23074F) in immunohistochemistry with formalin-fixed, paraffin-embedded tissues (IHC-P), we recommend antigen retrieval with either Sodium Citrate (Cat. No. 928502), or Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0). 

This antibody (clone W23074F) is not suitable for western blot, flow cytometry or immunocytochemistry.
