The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 2 µg/mL of the primary antibody for overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-mouse IgG for one hour at room temperature. Description

IFN-induced dsRNA-dependent serine/threonine-protein kinase (EIF2AK2) is an important component of the innate immune response to viral infection. The N-terminal of EIF2AK2 contains a double stranded RNA (dsRNA) binding domain, which binds dsRNA introduced to the host cells by viral infection, and activates a pro-apoptotic signal in the host cell. Recent studies have shown that activation of EIF2AK2 can lead to the accumulation of amyloid beta in Alzheimer disease patients, suggesting it plays a role in neurodegenerative disorders.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 0.5 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 0.5 - 10 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody reacts stronger with rat tissue samples than human cell line samples. Recommended WB usage for rat brain: 0.5 - 2 µg/mL; Recommended WB usage for human cell lines: 2 - 10 µg/mL
