Total lysates (15µg protein) from 293T, 293T/ERK1 knockout (KO) and 293T/ERK2 knockdown (KD) cells were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 1:1000 diluted (0.5µg/mL) purified anti-ERK1 antibody, clone W15133A (upper) or 1:3000 diluted anti-GAPDH (poly6314) antibody (lower). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-rat-IgG secondary antibody conjugated to HRP for the anti-ERK1 antibody, and a donkey anti-rabbit IgG Antibody conjugated to HRP for GAPDH. Lane M: Molecular weight ladder, M* indicates longer exposure. -
Total cell lysate from human Raji cells (lane 1), Jurkat cells (lane 2), and mouse A20 cells (lane 3) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal anti-ERK1 (clone W15133A) antibody. Proteins were visualized using an HRP Goat anti-rat IgG antibody and chemiluminescence detection. -
IHC staining using purified anti-Erk1 antibody (clone W15133A) on formalin-fixed paraffin-embedded human colon carcinoma tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05M) with Tween 20 (Cat. No. 925501), the tissue was incubated with 5 µg/mL of antibody overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-rat IgG antibody (Cat. No. 405416) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scalebar = 50 µM.
ERK1 is a member of mitogen-activated kinases (MAPKs) of serine/threonine protein kinases. ERK1 and ERK2 share 85% homology and both are involved in the MAP kinase signaling cascade. ERK1/2 can be activated by a range of extracellular stimuli, such as mitogen, growth factors, neurotransmitters, chemokines, and cytokines, through receptor tyrosine kinases (RTK), G protein-coupled receptors (GPCRs), or protein kinase C (PKC). Upon stimulation, ERK1/2 are phosphorylated by the upstream kinase MEK on residues Thr202 and Tyr204 and in turn phosphorylate many other downstream molecules that are involved in a range of cellular processes such as cell proliferation, differentiation, motility and cell death.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.2 - 1.0 µg per ml. For immunohistochemistry, a concentration range of 1 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
