Whole cell extracts (15 µg protein) from the indicated cell lines, BeWo (low expressor control), HaCaT, PC-3, MOLT-4, and NIH/3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-ETS1 antibody, clone W17042D, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular Weight marker. Predicted ETS1 expression was obtained from Human Protein Atlas.

Description

ETS1 is a transcription factor that regulates gene transcription of cytokine and chemokine genes. The expression of ETS1 is modulated by transcription factors and kinases. It is expressed in lymphoid organs where it may  play a role in cell differentiation, survival, and proliferation. The ETS1 gene encodes multiple proteins by  alternative splicing of mRNA, resulting in at least six distinctive isoforms; the major species are p51, p48, p42, and p39. In certain cancers, elevated ETS1 expression is a predictive marker of poor prognosis.

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

W17042D failed ICC testing; PC3 cells were fixed with 4% paraformaldehyde and tested using two  permeabilization methods (Methanol and Trition X-100). The clone failed ICC testing due to weak and non-specific staining of cytosol.

 

IP has not been tested on clone W17042D.
