    ICC staining of anti-Flotillin-1 antibody (clone W16108A) on Hela cells. The cells were transfected with plasmids carrying CRISPR gRNA against Flotillin-1, Cas9 nuclease, and GFP. Two days after transfection the cells were fixed with 4% PFA, permeabilized with 0.1% Triton X-100, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then stained with 5 µg/ml of the primary antibody overnight at 4°C, followed by incubation with Alexa Fluor® 594 goat anti-Rat IgG for one hour at room temperature. Nuclei were counterstained with DAPI. Arrowheads: transfected cells. Images were captured with a 60X objective. Scale bar: 25 µm.

Description

The flotillin family is composed of two highly homologous proteins, flotillin-1 and flotillin-2 that share ~50% amino acid sequence similarity. Flotillins are ubiquitously expressed, membrane-associated proteins, and were shown to be associated with lipid microdomains called membrane rafts. Flotillins are present in various cellular compartments, including the plasma membrane and endosomes, and they have been suggested to function in cellular signaling and membrane trafficking processes, including endocytosis and endosomal trafficking.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 5.0 µg per ml. For immunocytochemistry microscopy, a concentration range of 1.0-5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody specifically recognizes Flotillin-1 but not Flotillin-2, verified using WB.
